1. EpiCBE4max, a high-efficiency, one-step multiplex base editing system, was developed by integrating episomal elements with the CBE4max platform. 2. The epiCBE4max system achieved a simultaneous homozygous editing efficiency of 44.44% across five target genes in porcine fetal fibroblasts with undetectable off-target effects.
