A comprehensive quality control system for Chinese herbal medicine based on HPLC ratio quantitative fingerprinting and spectral quantized fingerprinting combined with antioxidant activity: A case of Siegesbeckiae Herba

A reliable quality control system is crucial to ensure the quality consistency, clinical efficacy, and safety of Chinese herbal medicines (CHMs) and their preparations. Herein, high performance liquid chromatography (HPLC) five-wavelength fusion fingerprint (FWFFP) and its ratio quantitative fingerprint (ROFP) were established, with the innovative ratio systematically quantitative fingerprint method (RSQFM) applied for quality assessment. The 22 batches of Siegesbeckiae Herbas (SHs) were classified into six Grades (0.845 < S mr < 0.980, 47.0 % < P mr < 112.3 %). Subsequently, the reliability of quality evaluation results was assessed using uncertainty parameters, with all values below the 0.2 threshold, confirming the robustness of this methodology. Additionally, the contents of four compounds were determined using both the standard curve method (SCM) and reference fingerprint quantitative method (RFPQM), revealing no significant difference between the two methods, thereby confirming the accuracy of RFPQM. Furthermore, ultraviolet (UV) and fourier transform infrared (FTIR) spectroscopic quantized fingerprint (QFP) were employed for structural characterization and qualitative-quantitative similarity evaluation. Then, the analytical results of HPLC, UV, and FTIR evaluated by RSQFM were integrated using an equal-weight fusion algorithm, all SHs were classified into five Grades (0.942 < S mr < 0.991, 78.6 % < P mr < 128.3 %).
