Epidermal Penetration Increases with Age and May Contribute to Systemic Inflammation

The skin is a major contributor to the age-associated rise in systemic inflammation, known as “inflammaging” (Yang et al, 2023; Ye et al, 2019). Previous research from our laboratory has shown that restoring epidermal barrier function in older mice, through application of a topical emollient, can normalize serum cytokine levels to those seen in young age (Hu et al, 2017; Man et al, 2015). This not only demonstrates that the skin is a major source of age-related systemic inflammation but also serves as proof of concept that this inflammation is amenable to treatment.
