Effects of Silencing BAK1 and BCL2 Gene Expression on Proliferation, Invasion and Metastasis of HCC HepG2 Cells

Objective: To investigate the effects of silencing BAK1 and BCL2 gene expression on \nproliferation, invasion and metastasis of hepatocellular carcinoma (HCC) HepG2 cells. \nMethods: 30 HCC HepG2 cells were randomly divided into groups and received the \ncorresponding treatments, namely, control group, NC-siRNA group, BAK1-siRNA group, \nBCL2-siRNA group and BAK1+BCL2 group, with 6 strains in each group. ThenqRT-PCR, \nCCK8, Transwell chamber invasion and scratch assay were used to detect the expression, \nproliferation, invasion and metastasis of BAK1 and BCL2 genes in HepG2 cells. Results: \nThe mRNA expression, cell proliferation rate, cell migration rate and cell invasion ability of \nBAK1 and BCL2 in HepG2 cells were lowest in the BAK1+BCL2 siRNA group, followed by \nBCL2-siRNA group, BAK1-siRNA group, NC-shRNA group and control group (P<0.05). The \nproliferation rate of HepG2 cells in the BAK1+BCL2 siRNA group decreased significantly \nwith time (P<0.05). Conclusion: Silencing the expression of BAK1 and BCL2 genes can \ninhibit the proliferation and invasion of HCC HepG2 cells and promote their apoptosis.
