Brentuximab Vedotin-Based Treatment in Patients with CD30-Positive Lymphoma: A Retrospective Analysis

Background: Brentuximab vedotin (BV) is an antibody-drug conjugate targeting CD30 which has been approved for the treatment of adult patients with CD30-positive lymphomas, specifically relapsed or refractory systemic anaplastic large cell lymphoma (sALCL) and classical Hodgkin lymphoma (cHL). This study presents a retrospective analysis of BV's efficacy and safety in CD30-positive lymphoma patients at our institution. Methods: This study included CD30-positive lymphoma patients treated with BV from July 2020 to April 2024. The primary endpoint was the overall response rate (ORR). The secondary endpoints were complete response (CR) rate, progression-free survival (PFS), overall survival (OS) and adverse events (AEs). Efficacy was assessed based on the 2014 Lugano Conference, and treatment-related AEs were evaluated according to CTCAE5.0. Results: A total of 66 patients were analyzed. The median age at diagnosis was 40 years (range: 15-79 years), with 43 (65.1%) being male. The patient cohort included 39 (59.1%) with cHL, 26 (39.4%) with peripheral T-cell lymphoma (PTCL), and 1 (1.5%) with primary mediastinal large B-cell lymphoma (PMBL). Two (3.0%) had previously undergone stem cell transplantation. The median number of prior chemotherapy lines was 0.5 (range: 0-7).
