The South China Cohort (SCC) is a population-based prospective cohort study aiming to gain a deeper understanding of the epidemiology, aetiology and risk factors of non-communicable diseases (NCDs) in southern China, a region with distinct climatic, dietary and lifestyle characteristics. A total of 116 520 adults aged 25–89 years were recruited from March 2018 to December 2020, from Guangdong, Fujian, Guangxi and Hainan Provinces. Passive follow-up will be conducted annually through linkages to established electronic registries, and 109 200 (93.7%) participants were successfully followed up in the first round; 20% of the participants are planned to be actively followed up every 5 years. The baseline dataset mainly included demographic and socioeconomic characteristics, personal and family health status, reproductive history (for women), lifestyle factors, environmental exposures and mental health. Additionally, physical examinations, laboratory tests and collections of biological samples were carried out. For the multi-omics sub-cohort of SCC, genomics, metagenomics, fungal internal transcribed spacer, untargeted metabolomics and proteomics were performed.
