Structure of the translating Neurospora ribosome arrested by cycloheximide

Significance The ribosome has the central role in the translation of RNA. The elongation function of most eukaryotic ribosomes is arrested by the drug cycloheximide (CHX). The site of CHX binding is known. In this work, we determine the structure of a translating ribosome of a filamentous fungus arrested by CHX. By doing so, the specific state of the ribosome arrested in translation is revealed. This work is significant because it provides a structural basis to establish CHX’s mechanism of action. It provides a model for a fungal ribosome and insights into the structure and function of eukaryotic ribosomes through comparative analyses of its features with those from other species.
