Functional Analysis of the Blackberry Sucrose Transporter Gene RuSUT2

Abstract In most plants, sucrose is the predominant form of carbohydrate for long-distance transport, and sucrose transporters (SUTs) mediate its transportation. In present study, we cloned a new sucrose transporter gene RuSUT2 from blackberry (Rubus spp.) cultivar Boysenberry fruit. The RuSUT2 cDNA contained an open reading frame (ORF) of 1986 nucleotides encoding a 631 amino acids protein. It was predicted that the protein exhibited 11 transmembrane spanning domain and the histidine residue (H) was conserved in the extracellular loop. Subcellular localization investigation indicated that RuSUT2 protein was localized in the plasma membrane. The phylogenetic comparisons revealed that RuSUT2 was a member of SUT2 clade and shared high identity with RcSUC3 in Rosa chinensis, FvSUC3 in Fragaria vesca, and PpSUT2 in Prunus persica. The quantitative real-time PCR (qRT-PCR) was performed to investigate the expression patterns of RuSUT2. The expression of RuSUT2 was detected in all test organs, and showed much higher expression in roots and leaves. During the fruit ripening stage, RuSUT2 was highly expressed at the 33 days after flowering (DAF). Overexpression of RuSUT2 enhanced sucrose content in the mature leaves of the transgenic tobacco. Our findings suggested that RuSUT2 has transport activity and could be involved in the sucrose transportation during blackberry plant growth and development.
