Effects of atorvastatin on mRNA expression of FKBP12 in vascular endothelial cell proliferation and the concentration of ET-1 in supernatant fluid of endothelial cell

Objective To evaluate the effect of atorvastatin on mRNA expression of FK506 binding protein 12 (FKBP12) in vascular endothelial cell(VEC) proliferation and the concentration of ET-1 in supernatant fluid of endothelial cell.Methods Rat vascular endothelial cells were cultured with enzyme digesting methods,and were identified by measuring their growth rate with invertmicroscope,by using immuocytochemical methods with factor Ⅷ antibody.Then the cells were treated with atorvastatin in different concentration.The proliferation of rat vascular endothelial cell was observed under an invert microscope.The expression of FKBP12 mRNA and the concentration of ET-1 in supernatant fluid of endothelial cell were detected in different culture periods.Results Atorvastatin markedly restrained the expressions of FKBP12 mRNA in VEC and the concentration of ET-1 in supernatant fluid of endothelial cell.A significantly positive correlation of them was indentified.The effect appeared dose dependence and time dependence.Conclusions Atorvastatin dose-dependently and time-dependently restrained the proliferation of rat VEC.As a result,the expression of FKBP12 mRNA and the concentration of ET-1 in supernatant fluid of endothelial cell are decreased too.
