The Effects of Pretreatment with Low-Frequency Repetitive Transcranial mMagnetic Stimulation on Expressions of Hippocampus GAD65 and NMDAR1 in Rats with Pilocarpine-Induced Seizures

Objective To study the effects of repetitive transcranial magnetic stimulation(rTMS)on Lithium-Pilocarpine induced seizures and its possible mechanisms.Methods Rats were randomly divided into the 0.5 rTMS group,sham stimulation group and control group.After every day for 2 weeks' consecutive corresponding stimulations,the acute epilepsy model was made by Lithium-Pilocarpine,and subsequently the latencies of seizures were observed,and the severity of seizure was quantitatively evaluated within 90 minutes after an intraperitoneal injection of Pilocarpine.Immunohistochemistry was used to observe the expressions of glutamic acid decar-boxylase65(GAD65)and N-methyl-D-aspartateR1(NMDAR1)in the rat hippocampus of all studied groups.Results Compared with sham stimulation group and control group,the latency of seizure became much longer,and the severity of seizure was much milder(P0.05 for both)in rTMS group;rTMS group had increased expression of GAD65,but decreased expression of NMDAR1(P0.05 for both)compared with the control group.There was no significant difference for expression of GAD65 and NMDAR1 between the sham stimulation group and control group(P0.05).Conclusions The rats pretreated with 0.5 Hz rTMS showed clear resistance to pilocarpine-induced seizure,and this effect might be produced by regulating the expression of GAD65 and NMDAR1 in the hippocampus.
