The culture conditions for the production of a new raw starch-digesting amylase which had been discovered in Chalara paradoxa were studied. Optimum culture medium consisted of 4.0% raw sago starch, 6.5% corn steep liquor, 0.7% beef extract, 0.3% NaCl and 0.08% FeSO4⋅7H2O. Optimum initial pH and optimum temperature were 4.5 and 30°C, respectively. This enzyme had higher affinity to raw starch compared with previously reported raw starch digesting amylases and hydrolyzed raw starch to glucose.
