Polyanionic Polymers in Bio- and Mucoadhesive Drug Delivery

Bio/Mucoadhesive drug delivery improves intimacy of contact between the drug delivery system and the absorptive surface and increases dosage form residence time. Bio/Mucoadhesive drug delivery systems adhere to the mucin-network and/or mucosal layer and remain in place until they dissolve or in the case of cross-linked polymers until the mucin or tissue layer replaces itself. Adhesive polymers can be neutral, anionic or cationic. On the basis of balance between adhesiveness and toxicity antonic polymers are preferred over neutral and cationic form. Bio/Mucoadhesion begins with establishment of intimate contact between the polymer and substrate. This is followed by bond formation and interpenetration/interdiffusion of polymer and substrate. Any factor that favors intimacy of contact, increases bond formation and enhances physical entanglement, will potentially increase bioadhesive strength.
