Study by X-ray powder diffraction and differential scanning calorimetry of two model drugs, phenothiazine and nifedipine, incorporated into lipid nanoparticles

Two drugs, phenothiazine and nifedipine, were incorporated into solid lipid nanospheres obtained by dispersion into cold water of warm O/W microemulsions, consisting of water, stearic acid, butanol and Tween 20®. Besides, mixtures of all the above components, excepting water, were submitted to the same thermal treatment. X-ray powder diffractometry (XRPD) and differential scanning calorimetry (DSC) showed that neither phenothiazine nor nifedipine did exhibit crystalline features in the drug-bearing nanospheres. On the contrary, they remain crystalline in the mixtures. It ensues that through our method of nanosphere preparation the initial crystalline state of the drugs changes to the amorphous one.
