A simplified method for the measurement of chemotaxis of polymorphonuclear leukocytes from human blood.

Abstract A simple reproducible method for the measurement of chemotaxis of polymorphonuclear leukocytes (PMN) with the use of small volumes of human blood is described. A cytocentrifuge is used to sediment the PMNs to a restricted area of a 3μ Millipore filter, which is placed in a standard tissue culture chamber. The method of cell counting allows a chemotactic index to be calculated. Normal values for this index have been set out and no correlation to age or sex has been found. The potential usefulness of this method is considered in the investigation of immunologic and metabolic disease states which have an increased frequency of infection and which may, in part, be related to abnormalities in the chemotaxis of PMNs.
