The covalent attachment of drugs to proteins and antibodies has been a subject of numerous investigations ( 1 ). These conjugated proteins are being evaluated as a means for delivering anticancer drugs to tumor sites ( 2 ) and are being used to elicit antibody responses from drugs in animals ( 3 – 5 ). The general analytical problem posed by these materials is the determination of the numbers of moles of materials of lower molecular weight (generally <2000 Da) relative to a mole of the materials of higher molecular weight (>10,000 Da), referred to as carriers, to which they are covalently bound. The molar ratio of the material of lower molecular weight to the carrier is referred to as the loading value. A related analytical problem is the determination of the distribution of the numbers of molecules of lower molecular weight covalently bound to carrier molecules. The relationship between the distribution and loading value is that the number average of the individual components making up the distribution is the average loading value.
