The effect of Bifidobacterium on CD4~+ CD25~+ Foxp3~+ Treg of blood and lymphonodi mesenterici in mice with ulcerative colitis

Objective To investigate the mechanism of Bifidobacteria treating the UC model mice through affecting the CD4+CD25+Foxp3+ regulatory T cells.Method Ulcerative colitis was induced in mice by oral administration of synthetic dextran sulfate sodium for 7 days,then followed with normal drinking water or Bifidobacterium for an additional 7 days.The mice were divided into normal control(NC) group,colitis model(MD) group,and the Bifidobacterium treated(BbT) group.The Treg percentages of CD4+CD25+Foxp3+/CD4+ were detected with flow cytometry.Result The DAI were increased significantly in MD group compared with NC group,and decreased in the BbT group than in MD group.The HDS were increased significantly in MD group compared with NC group,and decreased in the BbT group than in MD group.The blood and lymphonodi mesenterici of the CD4+CD25+Foxp3+/CD4+ Treg percentages were decreased significantly in MD group compared with NC group(P0.05),and increased in the BbT group than in MD group.Conclusion Bifidobacteria may play an important role in inhibiting the colonic inflammation in UC,by up-regulating CD4+ CD25+ Foxp3+ Treg expression.
