Expression of H1-calponin and clinical significance in gastric cancer

Objective To investigate the expression level of H1-calponin and analyze its relationship with matrix metalloproteinase 2(MMP-2) expression level as well as their clinical significance in gastric cancer.Methods Reverse transcription-polymerase chain reaction(RT-PCR) and Western blotting were performed to detect H1-calponin mRNA and protein levels in gastric cancer(n=20) or adjacent normal gastric mucosa tissues.Zymography and RT-PCR were used to detect the activity and mRNA levels of MMP-2,respectively,in gastric cancer or adjacent normal gastric mucosa tissues.Results In gray scale scanning with Western blotting,H1-calponin gene expression of gastric cancer was significantly lower than that of adjacent normal gastric mucosa tissues(0.98±0.28 vs 3.11±1.37,t=-6.833,P0.01).MMP-2 activity in gastric cancer cases was significantly higher than that in adjacent normal gastric mucosa tissues(3.48±1.25 vs 1.26±0.39,t=7.556,P0.01).A negative correlation was found between H1-calponin protein level and MMP-2 activity in gastric cancer tissues(r=-0.623,P0.01).Conclusion H1-calponin expression level is decreased in gastric cancer,which suggested the important role of H1-calponin in gastric cancer.
