Establishment of Efficient Regeneration System from Leaf Explants of Potato

The regeneration system of eight potato cultivars was studied with leafs as explants.The results showed that the optical callus induction medium was MS+6-BA 3 mg/L+NAA 0.3 mg/L+GA3 10 mg/L for Yunshu 501 and MS+6-BA 3 mg/L +NAA 0.5 mg/L +GA3 10 mg/L for Hui-2,and the induction rate reached to 100 % and 95.7 %,respectively.The optical shoot induction medium was found to be MS+6-BA 3mg/L +GA310 mg/L for Yunshu501 and MS+6-BA 3 mg/L+NAA 0.02 mg/L +GA3 10 mg/L for Hui-2,and the regeneration frequency reached to 86.3 % and 65.4 %,respectively.In contrast,no or very low callus regeneration was obtained from other cultivars,indicating that regeneration efficiency was strongly genotype-depended.
