Study of effects of carvedilol on transient outwardpotassium current (I_(to)) of ventricular myocytes of rats

【Objective】To explore the effects of carvedilol on transient outward potassium current (Ito) of ventric-ular cardiomyocytes of rats.【Methods】Rat cardiomyocytes were isolated with collagenase to acquire single ven-tricular myocytes. Ito current was recorded using whole- cell recording patch clamp technique. The influences ofcarvedilol on Ito were studied.【Results】Ito current density (pA/pF) was: (5.06±1.98), (3.91±0.66), respectively, when0.1 mM, 1.0 mM carvedilol was added to extracellular bath solution (P 0.05). Current- voltage relationship (I- V)curve was shifted downward.【Conclusions】Carvedilol has significant inhibitive effects on Ito. The inhibitive effectsare more prominent in higer concentrations than in lower concentratons.
