Altered Ribosomes in Antibiotic-Resistant Mutants of E. coli

Aminoglycoside antibiotics specifically affect protein synthesis in bacteria, and while the mechanism by which protein synthesis is stopped has not yet been completely elucidated (several steps in the protein synthetic process may be susceptible), the fact that mutational alteration of the ribosomes is the primary resistance mechanism has proved of great utility. Resistance to the aminoglycoside antibiotics in bacteria has been shown to produce altered 30 S ribosomal subunits, these mutations have allowed genetic mapping of the loci responsible for synthesis of the proteins of this subunit, and serve also as “markers” for ribosomal proteins.
