Abstract 1.1. The details are given of a modification of the Schoenheimer-Sperry method for the determination of free and total cholesterol in plasma or serum. 2.2. The results of the analyses of thirty-five normal human plasmas by this method are presented. 3.3. The constancy at which an individual can maintain the percentage of free to total cholesterol in his plasma is demonstrated by two extended studies on patients with cirrhosis of the liver. 4.4. The major points of departure of this method from the origipal are discussed.
