Laser Desorption/Ionization Mass Spectrometry of Proteins of Mass 100 000 to 250 000 Dalton

Irradiation of proteins in a nicotinic acid matrix with the 266‐nm light of a Nd‐YAG laser allowed the first determination of molecular masses up to 250 000 daltons. Since the peaks of multiply charged molecular ions and of oligomer ions can also be used in determination of molecular mass, this method attains a precision of ca. 0.1%. Its potential is demonstrated for β‐D‐galactosidase, glucose isomerase, and catalase.
