A new technique, “quantitative transplantation” of mammary glands in mice, is described in detail. Fifteen of 22 accurately located and measured segments of mammary gland from a 130-day-old hybrid male donor mouse (CBA × C57BL) were transplanted into the fourth mammary gland-free fat pads of 11 virgin female CBA × C57BL mice that subsequently mated and were observed through pregnancy. Fifteen of the transplanted mammary gland segments from a male donor regenerated fully developed mammary glands and lactated. They were indistinguishable, both macroscopically and microscopically, from the hosts' intact mammary glands at parturition. The mammary glands of the male mouse are similar to those of females in their growth potentiality and capability of initiation of lactation.
