Light-dependent Adenosine Triphosphatase in Chloroplasts

The presence or absence of an adenosine triphosphatase (ATPase) reaction in chloroplasts has been the subject of several recent investigations.Arnon (1) mentioned that the photosynthetic phosphorylation system seems to be rather free from ATP-hydrolyzing enzymes.Avron and Jagendorf (2, 3) showed that no significant ATPase activity could be found in chloroplasts under a variety of conditions.On the other hand, Baltscheffsky and Wessels (4, 5) reported significant ATPase activity in spinach chloroplasts in the dark.In addition, Petrack and Lipmann (6, 7) observed a light-dependent ATPase, induced by high concentrations of sulfhydryl reagents, in photophosphorylating particles from Anabena variabilis and in spinach chloroplasts.In view of the possible significance of an ATPase reaction in understanding the mechanism of photophosphorylation, we have reinvestigated the problem, utilizing the high sensitivity possible with highly labeled ATPs2 as substrate (8).
