Abstract The inactivation of the bacterial protease, subtilisin BPN', has been achieved with a substrate-like reagent, the bromomethyl ketone derived from benzyloxycarbonyl-l-phenylalanine. Loss of hydrolytic function accompanied the alkylation of N-3 of a single histidine residue. The bacterial enzyme has thus been shown to have a catalytic mechanism similar to that of mammalian pancreatic proteolytic enzymes.
